Hybridoma Production Protocol Payment for Phase One should be received before the project is initiated. Payment for Phase Two should be received before the frozen copies of the primary cultures and their associated culture medias are shipped. Due to the high fusion efficiency (~5000 to 10000 clones in ten 96 well plates) primary cultures should not be considered clonal. Moreover, artificially fused cells are initially genetically unstable. Such cells might expel a chromosome carrying a gene necessary for antibody production but retain their ability to divide, and can potentially overgrow antibody secreting cells. To avoid any loss of valuable clones by overgrowing of non-producers, we immediately subclone the 5 primary cultures chosen by the client before the frozen copies are made. If, in addition to the initial 5 primary clones chosen, the client wishes to expand more primary cultures, the client is expected to contact us within 2 days of receiving the ELISA screen (delays at this stage could result in the loss of an important hybridoma cell line). (€ 2600, approximately 6 weeks) If the subcloning of all 3 primary cultures is successful, the client receives 12 frozen cell line copies, 6 media samples, and monoclonal antibody isotyping results for 3 cell lines. Payment for Phase Three should be received before the subcloned cell lines and their associated culture medias are shipped.
Phase One: Immunization (€ 1000, approximately 60 days)
All shipments from ModiQuest will be shipped on dry ice. The fee for this service will be charged to the client.
Phase Two: Fusion (€ 2500, approximately 5 weeks)
Subcloning of additional primary cultures will be charged at € 800 per clone. As described above, if subcloning is successful, the client can expect 2 sister subclones, each frozen in double copies, culture media samples for each, and monoclonal antibody isotyping results.

(80 days, € 1000) Above immunisation protocol can be customized according to the clients specifications, and/or combined with the peptide antigen design and/or affinity purification protocols. ModiQuest has developed an efficient mix of anti-collagen antibodies for the generation of a collagen antibody induced arthritis (CAIA) mouse animal model.
Collagen-induced arthritis is an antibody mediated disease. Therefore, type II collagen specific antibodies can be used to induce arthritis in mice. Our mixture of type II collagen specific antibodies (2.6mg per animal) induces severe inflammation in the ankle and paws of DBA mice which resemble many aspects of arthritis. One of the biggest advantages of our monoclonal antibody-induced arthritis model is that severe arthritis is induced within 24-48 hours instead of the four weeks required to induce arthritis after immunization with type II collagen. ModiQuest Research recommends highly susceptible DBA/1 mice. Other strains may be used, however the amount of antibody cocktail required may vary. Thus, this model is ideal for rapidly screening therapeutics for the treatment of arthritis and to study the mechanisms involved in the development of arthritis.

All antibodies can be fully optimised towards specific diagnostic or therapeutic requirements. Using both gene synthesis to create novel expression constructs and antibodies, or adaptation of the large ModiQuest expression vector range to create or optimise antibodies for their specific applications.
ModiQuest has developed a sensitive Antibody Based Assay to test for Peptidyl Arginine Deiminase (PAD) activity (ABAP). The ABAP is an Enzyme Linked-Immuno-Sorbent Assay (ELISA) for the determination of PAD enzyme activity in cell and tissue lysates.
PAD enzymes are transcriptional co-regulators that catalyze the calcium dependent conversion of specific arginine residues present in for instance histones to citrulline. In diseases such as multiple sclerosis and rheumatoid arthritis (RA) these PAD enzymes seem to be dysregulated. For instance in RA these enzymes become active when they exit the cell in regions of joint inflammation where they deiminate arginines in proteins such as vimentin and fibrinogen. The level of PAD activity has been correlated with RA severity.
The ABAP test is also suitable for determining PAD activity of purified recombinant PAD enzymes used in various bioassays.
Via ModiQuest Research this kit is commecially available. The MQR-ABAP test is a powerful method to rapidly screen for PAD activity. Compared to currently used assays for PAD activity determination such as the DAMO BAEE conversion test the MQR-ABAP assay is more robust, and at least 500 times more sensitive.
The commercial availble ABAP kit consists of: 1) 96-well plate coated with Arginine containing peptides 2) A ModiQuest Research mouse PAD enzyme for activity calibration 3) a proprietary ModiQuest Research HRP-labelled monoclonal antibody specifically binding deiminated arginines. Bioreactor CL1000 Integra Antibody Production ModiQuest utilizes in vitro methods of antibody production as compared to the ascites fluid method, which needlessly destroys animals. The standard fee for one bioreactor production run includes:

At ModiQuest, we prefer to run an Integra CL1000 bioreactor for 6 weeks because this system allows the production of antibody at a reasonable price. Clients can expect a yield between 100-300 mg of their monoclonal antibody in approximately 8 weeks. Due to the variable secretion rates of hybridomas, the actual amount of antibody generated is dependent on the unique characteristics of each hybridoma.
One standard production run takes aproximately 8 weeks, € 2000*

Antigen specific B-cell selection offers a fast and efficient opportunity to obtain (recombinant) antibodies from various species (i.e. mouse, human, llama, chicken, rabbit etc.).
Difficult and rare antibody specificities can be generated using B-cell panning technology.

Depending on how you arrive in the Netherlands, this is how you can find the city of Nijmegen and the ModiQuest Facilities:
Parking:
All car parks in the vicinity are equipped with barriers and point-of-pay terminals. Regular parking cost € 0.80 per hour and a maximum of € 4 per day. After 18.00 and during weekends regular parking is free.
Plane Coming from the south (from Paris, Brussels, etc.), you will have to change trains at the first station in the Netherlands, Roosendaal. You take the inter-city train to Zwolle, and get off in Nijmegen. There is a train every thirty minutes, and the journey will take about an hour and a half
If you come to the Netherlands by air, you can take a train from Schiphol Airport to Duivendrecht (there is a train every thirty minutes), where you change trains. There are direct trains to Nijmegen Central Station every thirty minutes. The journey will take an hour and a half in total.
Train
If you are travelling by train (possibly preceded by a boat journey) from your own country, one of the following situations will apply:
Coming by boat from overseas (England, Ireland), you will probably arrive in Hoek van Holland. At Hoek van Holland you should take the train to Rotterdam Central Station, where you change trains for Breda. In Breda, board the train to Nijmegen. The train journey takes between 2 and a half and 3 hours. Coming from the southeast (Germany, Italy, Switzerland, Austria, etc.), the train will take you through Germany along the Rhine until you arrive in Arnhem. Trains between Arnhem and Nijmegen run every 10-15 minutes. It only takes a quarter of an hour to get from Arnhem to Nijmegen
Coming from the east (northern Germany, Denmark, boat-trains from Scandinavia, Poland, the New Independent States, etc.), the trains’ itinerary will include cities such as Hanover in Germany. Hengelo is the first station in the Netherlands. Do not get off there, but continue for half an hour until you get to Zutphen, where you change trains for Nijmegen. You should arrive in Nijmegen after approximately 40 minutes.
When travelling by train (www.ns.nl), keep three things in mind:
1. Always check at the railway station (or if possible at home) if the scheme mentioned above is still correct.
2. In the Netherlands, trains do not run all night. If you arrive on a very late international train, it is possible that you will not make it to Nijmegen that day. The latest possible arrival time ranges between 0.30 and 1.30 A.M.
3. Do not buy a ticket every time you change trains. In most cases you will be able to buy a ticket to Nijmegen in your hometown. If this is not the case, buy a ticket to Nijmegen at the first station in the Netherlands you enter, regardless of how many times you may have to change trains. This will save you much time (and money).
Train tickets and Pass Types
1. Singles and Day Returns (Enkele reis en Dagretour). It is cheaper to buy a Cheap Day Return than two singles. Your return journey must take place on the same day.
2. Day Travel Card (Dagkaart). This card allows you unlimited travel around the country in the space of one day. For € 4.50 extra, you can use other forms of public transport too, such as the bus, metro, or tram (this is called an OV Dagkaart).
Prices 3. Weekend Return (Weekendretour). A Weekend Return is the same as a Day Return, except that it is valid for a whole weekend. For the price of a day return you can make your journey between 7 P.M. Friday evening and 4 A.M. the following Monday morning.
4. Holland Rail Pass. This pass gives you unlimited access to all railways nation-wide for either three or five days, depending on the arrangement. You do not have to travel on consecutive days, but you must use your travel days within the period of one month. The Holland Rail Pass is available at railway ticket offices or at NS sales outlets abroad, upon presentation of your passport.
5. The Off-peak Discount Pass (Voordeel-urenkaart). If you travel after 9 A.M. Monday to Friday or on weekends, the Off-Peak Discount Pass (€ 49.00) enables you to travel within the Netherlands at a 40% discount. Off-Peak Discount Pass holders can travel with up to three people, who can enjoy the same 40% discount on the full fare. To obtain an Off-Peak Discount Pass, you must bring a passport photo (and the required amount of money) to the railway station ticket window, where you will fill out an application. After approx. three weeks you will receive the card at your home address. number 10 (Heyendaal shuttle) runs every 7 minutes.

Once you have arrived at Nijmegen Central Station, the easiest way to get to the ModiQuest facilities is by bus. It takes you 7 minutes to get to the ModiQuest facilities:
Get of at the HAN stop at the Kapittelweg, opposite of the Trigon building where ModiQuest B.V. is situated. `Strippenkaarten` are valid all over the Netherlands and can be used on any bus, tram or metro in the country. Strippenkaarten can be bought at the bus, tram or metro station, but the cheapest places to buy them are at the post office and the train station. On most buses it is the bus driver who stamps your ticket. Trams and some buses have devices that enable you to stamp the ticket yourself. Before you stamp your ticket, you need to know how many zones you will be crossing. Zone information is given on the information panels at bus, metro and tram stops.

ModiQuest: new targets and tools for diagnostics and therapeutics
COLLABORATIVE RESEARCH TO COMBAT RHEUMATOID ARTHRITIS
ModiQuest has entered into a research collaboration dedicated to the development of novel therapeutics to combat rheumatoid arthritis (RA). This progressive autoimmune disease leads to chronic inflammation and affects about one percent of the world`s population. ModiQuest will coordinate an extensive research program, aimed at the discovery of inhibitors of so-called PAD enzymes (peptidyl arginine deiminases), in which next to ModiQuest, two research groups from the Radboud University Nijmegen, a research group from the Leiden University Medical Center, and the fellow Dutch company Chiralix participate. Earlier research by the collaborators has identified the pivotal role of this class of enzymes in the onset and progression of RA. The consortium of these five partners has the necessary expertise to study each individual aspect of the complex project in great detail. ModiQuest, together with the research group of Dr. Ger Pruijn (Radboud University Nijmegen), will focus on the unraveling of biochemical aspects related to inflammatory processes that occur during rheumatoid arthritis. During these processes proteins are being modified and patients make antibodies against such modified proteins. Only in rheumatoid arthritis patients antibodies recognizing citrullinated proteins do occur. The citrullination of proteins is mediated by PAD enzymes, which only become activated when cells die. In rheumatoid arthritis inflammatory events in the joints of patients cause a chain reaction of protein citrullination by PAD enzymes followed by the generation of more antibodies that cause more inflammation resulting in more release of PAD enzymes leading to more citrullination and emerging into a chronic disease. The most promising class of PAD enzyme inhibitors are oligopeptides. These peptides should be functionalized in such a way that they are able to bind the active site of PAD, thereby blocking the enzyme from performing its natural function. To this end, amino acid building blocks are required, containing specific functionalities in the side chains. Chiralix, together with a team of researchers headed by Prof. Floris Rutjes (Radboud University Nijmegen) will explore the preparation of the amino acid-based compounds and derivatives.
