ModiQuest was founded by scientists of the Radboud University early 2004 as a private company. ModiQuest does focus on the development of novel diagnostics and therapeutics for autoimmune diseases.
ModiQuest’s primary line of business is the discovery and marketing of new antibodies, technologies and targets for (autoimmune) diagnostic and therapeutic applications.
ModiQuest applies various innovative technologies like differential proteomics for the identification of disease related protein modifications, automated high-throughput B-cell selection/culture, and highly efficient electrofusion methods for the production of mouse and human monoclonal antibodies.
Hybridoma Production Protocol Payment for Phase One should be received before the project is initiated. Payment for Phase Two should be received before the frozen copies of the primary cultures and their associated culture medias are shipped. Due to the high fusion efficiency (~5000 to 10000 clones in ten 96 well plates) primary cultures should not be considered clonal. Moreover, artificially fused cells are initially genetically unstable. Such cells might expel a chromosome carrying a gene necessary for antibody production but retain their ability to divide, and can potentially overgrow antibody secreting cells. To avoid any loss of valuable clones by overgrowing of non-producers, we immediately subclone the 5 primary cultures chosen by the client before the frozen copies are made. If, in addition to the initial 5 primary clones chosen, the client wishes to expand more primary cultures, the client is expected to contact us within 2 days of receiving the ELISA screen (delays at this stage could result in the loss of an important hybridoma cell line). (€ 2600, approximately 6 weeks) If the subcloning of all 3 primary cultures is successful, the client receives 12 frozen cell line copies, 6 media samples, and monoclonal antibody isotyping results for 3 cell lines. Payment for Phase Three should be received before the subcloned cell lines and their associated culture medias are shipped.
Phase One: Immunization (€ 1000, approximately 60 days)
All shipments from ModiQuest will be shipped on dry ice. The fee for this service will be charged to the client.
Phase Two: Fusion (€ 2500, approximately 5 weeks)
Subcloning of additional primary cultures will be charged at € 800 per clone. As described above, if subcloning is successful, the client can expect 2 sister subclones, each frozen in double copies, culture media samples for each, and monoclonal antibody isotyping results.
ModiQuest provides polyclonal Chicken antibody services
(80 days, € 1000) Above immunisation protocol can be customized according to the clients specifications, and/or combined with the peptide antigen design and/or affinity purification protocols. Please note that IgY cannot be purified using Prot A or Prot G. Payment for the polyclonal rabbit antibody services should be received before the project is initiated.

ModiQuest has developed an efficient mix of anti-collagen antibodies for the generation of a collagen antibody induced arthritis (CAIA) mouse animal model.
Collagen-induced arthritis is an antibody mediated disease. Therefore, type II collagen specific antibodies can be used to induce arthritis in mice. Our mixture of type II collagen specific antibodies (2.6mg per animal) induces severe inflammation in the ankle and paws of DBA mice which resemble many aspects of arthritis. One of the biggest advantages of our monoclonal antibody-induced arthritis model is that severe arthritis is induced within 24-48 hours instead of the four weeks required to induce arthritis after immunization with type II collagen. ModiQuest Research recommends highly susceptible DBA/1 mice. Other strains may be used, however the amount of antibody cocktail required may vary. Thus, this model is ideal for rapidly screening therapeutics for the treatment of arthritis and to study the mechanisms involved in the development of arthritis.

ModiQuest uses its unique Hi-tech facility to generate high producing primary cells and stable cell lines.
ModiQuest focusses on the generation of stable, high level, antibody producing cell lines (using AMAXA Nucleofection, high throughput ClonePix technology, and optimised proprietary vector systems).
Various Mouse and Human isotypes as well as Chicken based recombinant (chimeric) antibody producing cell lines can be generated very efficiently.
All antibodies can be fully optimised towards specific diagnostic or therapeutic requirements. Using both gene synthesis to create novel expression constructs and antibodies, or adaptation of the large ModiQuest expression vector range to create or optimise antibodies for their specific applications.
ModiQuest has developed a sensitive Antibody Based Assay to test for Peptidyl Arginine Deiminase (PAD) activity (ABAP). The ABAP is an Enzyme Linked-Immuno-Sorbent Assay (ELISA) for the determination of PAD enzyme activity in cell and tissue lysates.
PAD enzymes are transcriptional co-regulators that catalyze the calcium dependent conversion of specific arginine residues present in for instance histones to citrulline. In diseases such as multiple sclerosis and rheumatoid arthritis (RA) these PAD enzymes seem to be dysregulated. For instance in RA these enzymes become active when they exit the cell in regions of joint inflammation where they deiminate arginines in proteins such as vimentin and fibrinogen. The level of PAD activity has been correlated with RA severity.
The ABAP test is also suitable for determining PAD activity of purified recombinant PAD enzymes used in various bioassays.
Via ModiQuest Research this kit is commecially available. The MQR-ABAP test is a powerful method to rapidly screen for PAD activity. Compared to currently used assays for PAD activity determination such as the DAMO BAEE conversion test the MQR-ABAP assay is more robust, and at least 500 times more sensitive.

Bioreactor CL1000 Integra Antibody Production ModiQuest utilizes in vitro methods of antibody production as compared to the ascites fluid method, which needlessly destroys animals. The standard fee for one bioreactor production run includes:

At ModiQuest, we prefer to run an Integra CL1000 bioreactor for 6 weeks because this system allows the production of antibody at a reasonable price. Clients can expect a yield between 100-300 mg of their monoclonal antibody in approximately 8 weeks. Due to the variable secretion rates of hybridomas, the actual amount of antibody generated is dependent on the unique characteristics of each hybridoma.

In collaboration with Euro-Diagnostica B.V. this target was developed into a new highly sensitive diagnostic test for RA (anti-CCP test) which enables the early and specific detection of this disease. Depending on how you arrive in the Netherlands, this is how you can find the city of Nijmegen and the ModiQuest Facilities:
Parking:
All car parks in the vicinity are equipped with barriers and point-of-pay terminals. Regular parking cost € 0.80 per hour and a maximum of € 4 per day. After 18.00 and during weekends regular parking is free.
Plane Coming from the south (from Paris, Brussels, etc.), you will have to change trains at the first station in the Netherlands, Roosendaal. You take the inter-city train to Zwolle, and get off in Nijmegen. There is a train every thirty minutes, and the journey will take about an hour and a half
If you come to the Netherlands by air, you can take a train from Schiphol Airport to Duivendrecht (there is a train every thirty minutes), where you change trains. There are direct trains to Nijmegen Central Station every thirty minutes. The journey will take an hour and a half in total.
Train
If you are travelling by train (possibly preceded by a boat journey) from your own country, one of the following situations will apply:
Coming by boat from overseas (England, Ireland), you will probably arrive in Hoek van Holland. At Hoek van Holland you should take the train to Rotterdam Central Station, where you change trains for Breda. In Breda, board the train to Nijmegen. The train journey takes between 2 and a half and 3 hours. Coming from the southeast (Germany, Italy, Switzerland, Austria, etc.), the train will take you through Germany along the Rhine until you arrive in Arnhem. Trains between Arnhem and Nijmegen run every 10-15 minutes. It only takes a quarter of an hour to get from Arnhem to Nijmegen
Coming from the east (northern Germany, Denmark, boat-trains from Scandinavia, Poland, the New Independent States, etc.), the trains’ itinerary will include cities such as Hanover in Germany. Hengelo is the first station in the Netherlands. Do not get off there, but continue for half an hour until you get to Zutphen, where you change trains for Nijmegen. You should arrive in Nijmegen after approximately 40 minutes.
When travelling by train (www.ns.nl), keep three things in mind:
1. Always check at the railway station (or if possible at home) if the scheme mentioned above is still correct.
2. In the Netherlands, trains do not run all night. If you arrive on a very late international train, it is possible that you will not make it to Nijmegen that day. The latest possible arrival time ranges between 0.30 and 1.30 A.M.
3. Do not buy a ticket every time you change trains. In most cases you will be able to buy a ticket to Nijmegen in your hometown. If this is not the case, buy a ticket to Nijmegen at the first station in the Netherlands you enter, regardless of how many times you may have to change trains. This will save you much time (and money).
Train tickets and Pass Types
1. Singles and Day Returns (Enkele reis en Dagretour). It is cheaper to buy a Cheap Day Return than two singles. Your return journey must take place on the same day.
2. Day Travel Card (Dagkaart). This card allows you unlimited travel around the country in the space of one day. For € 4.50 extra, you can use other forms of public transport too, such as the bus, metro, or tram (this is called an OV Dagkaart).
Prices 3. Weekend Return (Weekendretour). A Weekend Return is the same as a Day Return, except that it is valid for a whole weekend. For the price of a day return you can make your journey between 7 P.M. Friday evening and 4 A.M. the following Monday morning.
4. Holland Rail Pass. This pass gives you unlimited access to all railways nation-wide for either three or five days, depending on the arrangement. You do not have to travel on consecutive days, but you must use your travel days within the period of one month. The Holland Rail Pass is available at railway ticket offices or at NS sales outlets abroad, upon presentation of your passport.
5. The Off-peak Discount Pass (Voordeel-urenkaart). If you travel after 9 A.M. Monday to Friday or on weekends, the Off-Peak Discount Pass (€ 49.00) enables you to travel within the Netherlands at a 40% discount. Off-Peak Discount Pass holders can travel with up to three people, who can enjoy the same 40% discount on the full fare. To obtain an Off-Peak Discount Pass, you must bring a passport photo (and the required amount of money) to the railway station ticket window, where you will fill out an application. After approx. three weeks you will receive the card at your home address. number 10 (Heyendaal shuttle) runs every 7 minutes.

Once you have arrived at Nijmegen Central Station, the easiest way to get to the ModiQuest facilities is by bus. It takes you 7 minutes to get to the ModiQuest facilities:
Get of at the HAN stop at the Kapittelweg, opposite of the Trigon building where ModiQuest B.V. is situated. `Strippenkaarten` are valid all over the Netherlands and can be used on any bus, tram or metro in the country. Strippenkaarten can be bought at the bus, tram or metro station, but the cheapest places to buy them are at the post office and the train station. On most buses it is the bus driver who stamps your ticket. Trams and some buses have devices that enable you to stamp the ticket yourself. Before you stamp your ticket, you need to know how many zones you will be crossing. Zone information is given on the information panels at bus, metro and tram stops.

If purchaser submits any acknowledgment of the order or other document that contains terms and conditions that are inconsistent with or in addition to the order or these terms and conditions, those additional or inconsistent terms are specifically rejected by ModiQuest and ModiQuest hereby objects to any such terms and conditions.
No variation of these terms and conditions will be binding upon ModiQuest unless agreed to in writing through non-electronic means and signed by an officer or other authorized representative of ModiQuest and shall be binding upon, inure to the benefit of, and be enforceable by, the parties hereto, and their respective heirs, personal representatives, successors and assigns.
2. Order Changes and Cancellations
Orders arising hereunder may be changed or amended only by written agreement signed by both purchaser and ModiQuest, setting forth the particular changes to be made and the effect, if any, of such changes on the price and time of delivery.
Purchaser may cancel an order only by providing written notice to ModiQuest at least 14 calendar days prior to the scheduled products delivery date or start of the services. Cancellation of an order during a project shall result in a charge of hundred (100) percent of the gross sales price of the original order.
3. Delivery and Acceptance
ModiQuest will use its reasonable commercial efforts to fulfill purchaser’s orders pursuant to any agreement for the supply of products.
All products ordered by purchaser pursuant to this Agreement shall be delivered F.O.B. ModiQuest` shipping point in the . Products shall be deemed accepted upon delivery. Purchaser may revoke acceptance of any shipment of products which are not in compliance with the specifications as provided under warranty herein. In order to revoke acceptance, purchaser must pay for the shipment as provided in Section 6 and comply with the provisions of Section 9.
ModiQuest will provide any advice and perform all other services, if any, to the best of its knowledge and ability and in accordance with professional standards.
4. Allocation of Goods
If ModiQuest is unable for any reason to supply the total demands for non-custom products specified in Purchaser`s order, ModiQuest may allocate its available supply among any or all Purchasers on such basis as ModiQuest may deem fair and practical, without liability for any failure of performance which may result therefrom
5. Prices and Taxes
Unless otherwise stated, prices are without engagement and may be altered by ModiQuest to those ruling at the date of despatch. Should the prices be increased in consequence thereof, then upon receipt of our notification of such price-increase the purchaser shall have the right to cancel the order in respect of the products which have not yet been despatched.
All taxes, duties, levies and similar expenses, which are or become due in connection with ModiQuest’s quotation, contract or any order resulting therefrom and the carrying out thereof are for ModiQuest’s account as far as they are due in the supplier`s country, and for the account of the purchaser as far as they are due in the purchaser`s country or in such other country or countries for which the products are destined or in which any services will be performed, irrespective of which party (including its representatives /employees) will be liable to pay such taxes.
6. Payment
Purchaser shall pay all invoices for products ordered by and delivered to purchaser without any deductions, discounts, set off or debt settlement within thirty (30) days of the receipt thereof in the official European currency. If an invoice balance is overdue, without waiving any other rights and remedies at law or under this Agreement, ModiQuest may (a) refuse to accept additional orders; (b) refuse to ship ordered products; and/or (c) seek collection from purchaser, including all legal fees and other costs of collection.
If the financial condition of purchaser results in the insecurity of ModiQuest, in its sole discretion, as to the ultimate collectability of the purchase price, ModiQuest may, without notice to purchaser, delay or postpone the delivery of the products; and ModiQuest, at its option, may change the terms of payment to payment in full or in part prior to shipment of the entire undelivered balance of said products.
In the event of default by purchaser in the payment of the purchase price or otherwise, purchaser agrees to pay the balance then due to ModiQuest on demand. Purchaser also agrees to pay the statutory interest from the due date until the date on which payment is made in full, and all costs, including, but not limited to, reasonable attorney and accounting fees and other expenses of collection resulting from any default by purchaser in any of the terms hereof.
7. Retention of title
Title to and ownership of products delivered shall be retained by ModiQuest until ModiQuest has received payment in full. Until the moment of full payment of all products delivered, the purchaser shall not be authorised to give or permit third parties the use of the Products, to rent or pledge, to transfer the title or to alienate or encumber same in any other way.
8. Limited Warranty
ModiQuest warrants to purchaser for a period of thirty (30) days from the date of delivery, that the products, when shipped to purchaser by ModiQuest, shall conform in all material respects to the specifications of ModiQuest, as provided on ModiQuest` web-site or in other literature provided to purchaser by ModiQuest; provided however, that the products must be stored under the prescribed conditions during such warranty period.
ModiQuest’s sole liability under this guarantee shall be to replace such parts or products as have proved to not conform to specifications as set out here above or, at ModiQuest’s option, repair such parts or products or have them repaired at ModiQuest’s order, always free of charge, provided that (i) ModiQuest is informed by the purchaser in writing (including telefax) within fourteen days after the defect(s) have revealed themselves (ii) upon receipt of shipping instructions from ModiQuest, purchaser shall return to ModiQuest, at ModiQuest’s cost, all products allegedly not conforming to the specifications as warranted; provided, however, in the event that it is subsequently determined that such products do in fact conform to the specifications in all material respects, purchaser shall reimburse ModiQuest for all such shipping costs incurred by ModiQuest, (iii) such defective parts and products shall become ModiQuest’s property as soon as they have been replaced. The warranty does not cover damage sustained by normal wear and tear or any damage arising in consequence of negligence or improper handling or use of the Products or parts thereof, or of improper installation or of maintenance by unauthorized persons, or of improper storage in the event of the products wholly or partly being stored by the purchaser previous to installation or use
Except for the foregoing warranty, ModiQuest does not warrant the merchantability or fitness for a particular purpose of the products or the performance or noninfringement thereof, does not make and hereby expressly disclaims any warranty, express or implied, with respect to the products, specifications, support, services or anything else relating to the products and does not make any warranty to purchaser, purchaser`s customers or their agents concerning the products and services.
The services rendered by ModiQuest and the result of an antibody project is dependent on the antigenicity of the material provided by the purchaser. ModiQuest does not guarantee that the antibodies developed apply in a particular technique or that the material supplied by the customer is immunogenic.
In addition, ModiQuest does not warrant that the use or sale of the products delivered hereunder will not infringe intellectual property rights covering the product itself or the use thereof in combination with other products or in the operation of any process.
9. Limited Liability
Purchaser acknowledges and agrees that ModiQuest` sole liability and purchaser`s sole and exclusive remedy hereunder for any breach of contract, or based on tort or other concept of law or theory shall not exceed the amount paid by purchaser for such products or services under the order, or in case of a specific fee paid for certain advise, the amount of said fee. Such limitations on ModiQuest` liability hereunder shall apply even if ModiQuest` liability is due in whole or in part to its own negligence. In order to obtain said product remedies, purchaser shall afford ModiQuest prompt and reasonable opportunity to inspect all products as to which any claim is made that such products do not conform to the warranties provided herein. At ModiQuest’ option, purchaser shall allow ModiQuest to take control of such products and direct their disposal.
In no event shall ModiQuest or its affiliates or their respective representatives be liable to purchaser or its affiliates , whether based in contract, tort, warranty or any other legal or equitable grounds, for any loss of the income, profit or savings or cost of capital of purchaser or its affiliates, for any indirect or consequential damages resulting from or relating to the order or the products delivered or services provided hereunder, even if ModiQuest has been advised of the possibility of such damages.
10. Purchaser`s Use of Products
ModiQuest` products are intended primarily for laboratory research purposes and, unless otherwise stated on product labels, on ModiQuest` web-site or in other literature furnished to purchaser by ModiQuest, are not to be used for any other purposes, including but not limited to, in vitro diagnostic purposes, foods, drugs, medical devices or cosmetics for humans or animals.
Purchaser acknowledges that the products have not been tested by ModiQuest for safety and efficacy in food, drug, medical device, cosmetic, commercial or any other use, unless otherwise stated in ModiQuest` literature furnished to Purchaser. Purchaser realizes that, since ModiQuest` products are, unless otherwise stated, intended primarily for research purposes, they may not be on the United States Toxic Substances Control Act (TSCA) inventory or similar inventory in any other country. Purchaser assumes responsibility to assure that the products purchased from ModiQuest are approved for use under the law of the state or country of its residence, if applicable. Purchaser has the responsibility to verify the hazards and to conduct any further research necessary to learn the hazards involved in using products purchased from ModiQuest. Purchaser agrees to comply with instructions, if any, furnished by ModiQuest relating to the use of the products and not misuse the products in any manner. No products purchased from ModiQuest shall, unless otherwise stated, be considered to be foods, drugs, medical devices or cosmetics.
11. Intellectual Property Rights
By entering into each and any contract, the purchaser acknowledges that nothing shall be construed to give the purchaser either during or after the duration of the contract any right, title or interest in the intellectual property rights with regard to the products.
All rights in inventions, all copyrights and all other intellectual or industrial property rights in work or resulting from work done by or one behalf of the purchaser pursuant to any contract shall vest in ModiQuest. The purchaser agrees to render its full assistance and cooperation to ModiQuest in obtaining and enjoying such rights.
Antibodies produced, and the reagents subsequently supplied by ModiQuest to the purchaser and fully paid for, shall become the property of the purchaser unless agreed otherwise.
In the event purchaser requires ModiQuest to perform services concerning materials or processes owned by the purchaser or third parties or unreveiled sources, the purchaser shall grant ModiQuest a license under its intellectual property rights to perform same and hold harmless and indemnify ModiQuest from and against any third party claim whether as a consequence of infringement of said third party’s intellectual propert rights or for any other reason not attributable to ModiQuest.

Purchaser represents and warrants that it shall use all products ordered in a lawful manner. Purchaser shall defend ModiQuest, its employees, agents, affiliates and Contractors (the "ModiQuest Indemnitees"), and shall hold the ModiQuest Indemnitees harmless from and against all suits, actions, or proceedings, at law or in equity, and from all claims, costs, damages, losses and expenses (including, without limitation, attorney`s fees, consultants` fees, experts` fees) of third parties that are related to or in connection with (1) the product or product information, if applicable, being wrongfully disclosed by purchaser hereunder, (2) infringement, misappropriation, and/or conversion which are the direct result of ModiQuest` possession and/or use of such products or product information disclosed by purchaser, if applicable, or (3) the death or bodily injury of any third party or the damage, loss or destruction of any tangible personal or real property arising from or related to purchaser`s use of the products, or its manufacture or sale of any products or utilizing the products.
Delay in performance or non-performance of any obligation contained herein, other than purchaser`s obligation to pay, shall be excused to the extent such failure or non-performance is caused by force majeure. For purposes of these Terms and Conditions, force majeure shall mean any cause or event preventing performance of an obligation under the order which is beyond the reasonable control of ModiQuest or purchaser, as the case may be, including without limitation, fire, flood, power shortage, mechanical breakdown, sabotage, shipwreck, embargo, explosion, strike or other labor trouble, accident, riot, acts of governmental authority (including, without limitation, act based on laws or regulations now in existence as well as those enacted in the future), acts of God, and other events or conditions beyond the reasonable control of the affected party. If purchaser or ModiQuest is affected by force majeure, the party affected shall promptly provide notice to the other party, explaining in detail the full particulars and the expected duration thereof. The affected party shall use its commercially reasonable efforts to remedy the interruption or delay if it is reasonably capable of being remedied. In the event a force majeure situation extends for more than thirty (30) days, the order may be terminated without any liability by either party upon written notice thereof to the other. In the event of a force majeure compelling ModiQuest to allocate production and deliveries of products, ModiQuest may allocate its available supply of products among ModiQuest` customers (including purchaser) and ModiQuest` internal uses in such manner as ModiQuest deems fair and reasonable. Such allocation shall not be deemed a breach of this Agreement.
14. Governing Law; Venue
This Agreement shall be subject to and construed in accordance with Dutch law. The United Nations Convention on Contracts for the International Sale of Goods shall not be applicable
The exclusive venue for proceedings arising under this Agreement shall be a court of competent jurisdiction in

ModiQuest: new targets and tools for diagnostics and therapeutics
COLLABORATIVE RESEARCH TO COMBAT RHEUMATOID ARTHRITIS
ModiQuest has entered into a research collaboration dedicated to the development of novel therapeutics to combat rheumatoid arthritis (RA). This progressive autoimmune disease leads to chronic inflammation and affects about one percent of the world`s population. ModiQuest will coordinate an extensive research program, aimed at the discovery of inhibitors of so-called PAD enzymes (peptidyl arginine deiminases), in which next to ModiQuest, two research groups from the Radboud University Nijmegen, a research group from the Leiden University Medical Center, and the fellow Dutch company Chiralix participate. Earlier research by the collaborators has identified the pivotal role of this class of enzymes in the onset and progression of RA. The consortium of these five partners has the necessary expertise to study each individual aspect of the complex project in great detail. ModiQuest, together with the research group of Dr. Ger Pruijn (Radboud University Nijmegen), will focus on the unraveling of biochemical aspects related to inflammatory processes that occur during rheumatoid arthritis. During these processes proteins are being modified and patients make antibodies against such modified proteins. Only in rheumatoid arthritis patients antibodies recognizing citrullinated proteins do occur. The citrullination of proteins is mediated by PAD enzymes, which only become activated when cells die. In rheumatoid arthritis inflammatory events in the joints of patients cause a chain reaction of protein citrullination by PAD enzymes followed by the generation of more antibodies that cause more inflammation resulting in more release of PAD enzymes leading to more citrullination and emerging into a chronic disease. The most promising class of PAD enzyme inhibitors are oligopeptides. These peptides should be functionalized in such a way that they are able to bind the active site of PAD, thereby blocking the enzyme from performing its natural function. To this end, amino acid building blocks are required, containing specific functionalities in the side chains. Chiralix, together with a team of researchers headed by Prof. Floris Rutjes (Radboud University Nijmegen) will explore the preparation of the amino acid-based compounds and derivatives.
